It is suitable for high-density suspension cultures of various Chinese Hamster Ovary (CHO) cell subtypes, including CHO-K1, CHO DG44, and CHO-S cells, enabling high-level expression of recombinant proteins and antibodies. Pricella® hMSCs Serum-Free Medium is specifically formulated for the growth and expansion of human mesenchymal stem cells (hMSCs). Compared to traditional serum-supplemented media (DMEM + 10% FBS), Pricella® hMSCs Serum-Free Medium better promotes hMSCs growth and enhances consistency. Using this SFM, hMSCs can be expanded for over five passages while retaining their mesodermal differentiation potential across three lineages- osteocytes, chondrocytes, and adipocytes. Each batch of hMSCs Serum-Free Medium is validated using hMSCs to ensure consistent performance, and hMSCs cultured in serum-supplemented media can transition directly to Pricella® hMSCs Serum-Free Medium with little to no adaptation required. Additionally, this SFM can also be used for the direct expansion of hMSCs from primary human bone marrow.

hMSCs

hMSCs were cultured and expanded using Pricella® hMSCs Serum-Free Medium (Cat. No.: CM-SC01).Flow cytometry was used to assess the expression of cell surface antigens. The results showed negative expression for CD19 (0.35%), CD34 (1.23%), and CD45 (1.08%), while positive expression was observed for CD90 (99.94%) and CD105 (97.02%).

Serum-Free Adaptation Process

Step 1: Seed cells in the original medium (serum-supplemented medium) and, once they enter the exponential growth phase, initiate serum-free adaptation.

Step 2: Culture cells in a medium composed of 75% original medium and 25% SFM.

Step 3: Culture cells in a medium composed of 50% original medium and 50% SFM.

Step 4: Culture cells in a medium composed of 25% original medium and 75% SFM.

Step 5: Culture cells in 100% SFM.

Tips: If cells cannot adapt smoothly from 75% to 100% SFM, consider adding a step with 10% original medium and 90% SFM. For each step, it is recommended to maintain the respective ratios and passage cells for 2-3 generations. If adaptation is successful, the number of passages at each step may be reduced based on actual conditions.
